Multiplication of Dendranthema zawadskii ssp. latilobum var. leiophyllum through Multiple Shoot Induction from Rhizome-Derived Shoot Tip Culture

|
Direct multiple shoots were induced in shoot tips derived from in vivo rhizomal buds of Dendranthema zawadskii ssp. latilobum var. leiophyllum on Murashige and Skoog (MS) medium supplemented with 6-benzylaminopurine (BAP) alone at 0.5-3.0 mg¡¤L^(-1) or combinations of 0.5-3.0 mg¡¤L^(-1) BAP, and 0-0.5 mg¡¤L^(-1) ¥á naphthaleneacetic acid (NAA). A combination of 0.5 mg¡¤L^(-1) BAP.0.01 mg¡¤L^(-1) NAA, and 30 g¡¤L^(-1) sucrose was optimal for multiple shoot formation (45 shoots per shoot tip) without roots within 4 weeks. Subculture of these shoots on the same medium resulted in similar reponses in multiple shoot formation within 4 weeks. Of the four basal media (1/2MS, MS, 2MS, B_(5)), MS basic medium was most effective for inducing multiple shoot formation. Adventitious shoots (more than 40 per multiple shoots) were formed continuously on MS basal medium supplemented with 0.5 mg¡¤L^(-1) BAP and 0.01 mg¡¤L^(-1) NAA through four generations of subculture. Compact plantlets were induced on MS medium containing with 0.5 mg¡¤L^(-1) paclobutrazol in vitro.
